Özet
Clonal rootstocks are commonly preferred due to their abilities to obtain early fructification and
maximum yield per unit area. In vitro techniques enable rapid and intensive propagation of rootstocks
in laboratory conditions. In this study, propagation possibility of Weiroot 158, an important clonal
rootstock used in cherry cultivation, was investigated using tissue culture technique. Shoot tips were
used as explants and Murashige-Skoog (MS) was used as the nutrient media (30 g L-1 sucrose and 7
g L-1 agar). The shoot tips were placed in the temperature and light controlled culture room after
planting in the nutrient medium. Samples were taken for subculture once in a month. Number of
explants, leaves and sprouting were measured every 20 days. Four different MS media were tested
during the shoot multiplication phase. The best shoot proliferation was obtained in a media
supplemented with 4.4 μM BAP+0.49 μM IBA+0.29 μM GA3. In addition, five different MS media
were tested during the rooting phase. The results showed that the best rooting was obtained in nutrient
media containing 1/2 MS + 2 mg/L IBA.
