TRNL-F bölgesi analizi yoluyla bazı nar (Punica granatum L.) genotipleri arasındaki genetik polimorfizmin belirlenmesi

Özet
Çalışmamızda 24 adet nar genotipinin TrnL-f bölgesi analizi yoluyla genetik akrabalıkları araştırılmıştır. Mardin yöresine ait 18 tane yerel genotipin yer aldığı çalışmada ayrıca Siirt yöresine ait Siirt-Çekirdeksiz adlı genotip ve beş adet tescilli nar çeşidi (Zivzik, Esin, Yılmaz, Hicaz ve Onur) bulunmaktadır. Nar genotipleri ve çeşitlerinin kloroplast genomuna ait TrnL-F bölgesi kullanılarak genetik karakterizasyonları belirlenmiştir. Nar genotiplerinde TrnL-F bölgesine ait primer kullanılarak DNA bölgeleri çoğaltılmış ve PCR sonuçlarına göre DNA dizi analizleri yapılmıştır. Çalışılan nar genotiplerinin akrabalık ilişkilerini belirlemek amacıyla filogenetik ağaçlar çizilmiştir. Maximum Likelihood metodu ve Neighbour-Joining metodu kullanılarak oluşturulan iki dendrogramda birbiriyle uyumlu benzer sonuçlar elde edilmiştir. Her iki dendogramda oluşan alt gruplara 15 nolu örnek dışarıdan bağlanmakta ve diğer genotiplere en uzak akraba olan genotip olduğu görülmektedir. Ancak, Maximum Likelihood analizi genotiplerin birbiri olan yakın akrabalık durumunu daha net biçimde ortaya koyarken, Neighbour-Joining analizi sonucunda elde edilen dendogramda 15 nolu örnek hariç diğerlerinin tek bir grup oluşturduğu görülmektedir.
In our study, genetic relationships of 24 pomegranate genotypes was investigated by TrnL-f region analysis. In the study, which included 18 local genotypes from the Mardin region, there were also the Siirt-Çekirdeksiz genotype belonging to the Siirt region and five registered pomegranate varieties (Zivzik, Esin, Yılmaz, Hicaz and Onur). Genetic characterization of pomegranate genotypes and cultivars were determined using the TrnL-F region of the chloroplast genome. In pomegranate genotypes, DNA regions were amplified by using the primer of TrnL-F region and DNA sequence analyzes were performed according to PCR results. Phylogenetic trees were drawn to determine the kinship relationships of the studied pomegranate genotypes. Similar results were obtained in two dendrograms created using the Maximum Likelihood method and the Neighbor-Joining method. Sample 15 is externally attached to the subgroups formed in both dendograms and it is seen that it is the genotype that is the most distantly related to other genotypes. However, while Maximum Likelihood analysis reveals the close relationship of genotypes to each other more clearly, in the dendogram obtained as a result of Neighbor-Joining analysis, it is seen that all the others, except for sample 15, form a single group.
