Öğe EFFECTS OF CEFAN MELON (CUCUMIS MELO L.) SEED EXTRACTS ON HUMAN ERYTHROCYTE CARBONIC ANHYDRASE I-II ENZYMES(Corvinus Univ Budapest, 2019) Akkemik, E.; Aybek, A.; Felek, ICarbonic anhydrases (CAs, E.C.4.2.1.1) are an important metalloenzyme family that catalyzes reversible CO2 hydration and HCO3- dehydration in organisms. CA inhibitors can be used in the treatment of many diseases such as glaucoma, epilepsy, obesity, and cancer. This study aimed to determine the effects of oil and methanol extracts of Cefan melon (Cucumis melo L.) seed (CMS) on CA enzyme activity. While both extracts activated hCA-I isoenzyme, they inhibited hCA-II. In order to determine the active substance causing inhibition or activation in CMS, essential oil content, phenolic substance profile and macro-micronutrient elements were investigated. Dominant fatty acids were identified as linoleic and oleic acid, while the dominant phenolic compound was gallic acid. Besides, it was found that CMS contained high concentrations of Ca, K and Na, and was rich in Se. Results demonstrated that essential oil and phenolic compounds in Cefan melon have potential applications as alternative natural products for pharmaceutical industries. In addition, Cefan melon seed has a wide range of applications for food and cosmetic industries based on the research into the fatty acid, phenolic content, and nutrient elements.Öğe Microwave Assisted Synthesis of Benzo-Azacrown Ethers and In Vitro Inhibition Studies on hCA I-II(Maik Nauka/Interperiodica/Springer, 2023) Sapmaz, A.; Calisir, Ue; Akkemik, E.; Cicek, B.In this study, macro-ringed benzo-aza-oxa-crown ethers were synthesized by a cyclization reaction between 2,2 '-dithiodianiline and mono/di/tri/tetraethylene glycol dichloride/dibromide with Cs2CO3 as a catalyst under nitrogen atmosphere by the S(N)2 mechanism using the microwave synthesis method, which is one of the green chemistry methods. Structural characterizations of the original 2,2 '-dithiodibenzo crown ether derivatives were confirmed with melting point, FT-IR, H-1 NMR, C-13 NMR, LC-MS/MS, elemental analysis, UV and fluorescence spectroscopy methods. In addition, the inhibition effects of these synthesized four crown ethers on human erythrocyte carbonic anhydrase (hCA I-II) isoenzyme activities were investigated. The synthesized compounds include aromatic benzene rings, nitrogen, sulfur, and oxygen donor atoms. The effects of aromaticity, S,S '-disulfide bond, nitrogen and oxygen donor atoms on the activity of carbonic anhydrase isoenzyme have been studied. (1,8,11,18)-Tetraaza[2,3-6,7-12,13-16,17]tetrabenzo-(4,5,14,15)-tetrathiocycloeicosane showed the highest inhibition effect on hCA I (K-i = 0.353 +/- 0.154 mu M) and hCA II (K-i = 1.588 +/- 0.998 mu M) enzyme.Öğe The impact of Tip60 gene on antioxidant system(Wiley-Blackwell, 2016) Kocpinar, E.
